The FN III region contains an alternatively spliced domain, which results in various Tenascin-C isoforms.

In tissues, Tenascin-C subunits assemble into trimers through interactions at the N-terminal coiled domain and further form hexamers via disulfide bonds. Low molecular weight forms of Tenascin-C are commonly found in normal tissues, whereas high molecular weight variants are significantly expressed in pathological conditions, including various cancers.

Significance: High molecular weight Tenascin-C forms, particularly those with the alternatively spliced Domain B, are associated with pathological processes, including cancer.
