Tecan IBL Dentin Matrix Protein 1 (DMP1)

General Characteristics:

  • Family: DMP1 is a member of the Small Integrin-Binding Ligand, N-linked Glycoprotein (SIBLING) family.
  • Size and Structure: Synthesized as a 105 kDa proprotein, which is then cleaved into a 37 kDa N-terminal fragment and a 57 kDa C-terminal fragment. It contains multiple acidic domains that interact with calcium.

Biological Functions:

  • Bone Mineralization: DMP1 is a crucial component of the extracellular matrix in osteocytes and plays a significant role in bone mineralization. Its acidic domains facilitate calcium binding, contributing to the mineralization process.
  • Osteocyte Function: Unlike other matrix proteins like Osteopontin, Osteocalcin, and Bone Sialoprotein, which are produced by osteoblasts, DMP1 is specifically expressed by osteocytes. Osteocytes are involved in regulating bone remodeling by controlling the activity of osteoclasts and osteoblasts. They also function as endocrine cells by secreting Fibroblast Growth Factor 23 (FGF-23), which regulates phosphate metabolism.
