APRIL (A Proliferation-Inducing Ligand) Overview:
- Family: APRIL is a member of the Tumor Necrosis Factor (TNF) family.
- Expression: It is highly expressed in various tumors and exhibits low expression in normal cells.
- Receptors: APRIL shares receptors TACI (Transmembrane Activator and CAML Interactor) and BCMA (B-cell Maturation Antigen) with BlyS/BAFF (B-lymphocyte Stimulator/BAFF), another TNF homolog. Both TACI and BCMA are associated with autoimmune diseases and cancer.
Functions:
- T Cell Survival and Proliferation:
- APRIL plays a role in T-independent type II antigen responses.
- It supports T cell survival and proliferation.
- Non-Lymphoid Cells:
- APRIL has the capacity to induce proliferation and survival in non-lymphoid cells.
- Inflammatory Conditions:
- In patients with inflammatory arthritis, APRIL is locally produced in affected joints.
- Biologically active heterotrimers of BlyS/BAFF and APRIL are found in systemic immune-based rheumatic diseases.
Interaction with BCMA:
- Soluble BCMA Receptor:
- A soluble form of the high-affinity BCMA receptor can inhibit APRIL’s proliferative activity in vitro.
- This inhibition reduces tumor cell proliferation.
- APRIL Overexpression:
- Cells transfected with APRIL show increased tumor growth, indicating its role in promoting tumor cell proliferation.
